Stress and anxiety are among the most commonly reported health concerns worldwide, and interest in plant- and fungi-based approaches to supporting psychological wellbeing has grown substantially in recent years. Among the candidates receiving the most research attention are functional mushrooms, several of which demonstrate properties that may influence the biological pathways involved in the stress response.
This article examines what current evidence says about functional mushrooms and their potential effects on stress, anxiety, and related physiological markers such as cortisol and inflammatory cytokines.
The Biology of Stress and Why Mushrooms May Be Relevant
The stress response involves multiple interconnected systems, including the hypothalamic-pituitary-adrenal (HPA) axis, the sympathetic nervous system, and inflammatory signaling pathways. Chronic activation of these systems is associated with elevated cortisol, increased adrenocorticotropic hormone (ACTH), and elevated pro-inflammatory markers such as C-reactive protein (CRP) and interleukin-6 (IL-6).
Functional mushrooms contain bioactive compounds, including triterpenes, beta-glucans, polysaccharides, and, in the case of Lion’s Mane, unique compounds called hericenones and erinacines. Several of these compounds have been investigated for their potential effects on neurological function, immune modulation, and stress biomarkers.
Lion’s Mane and Mood: Clinical Evidence
Hericium erinaceus, commonly known as Lion’s Mane, has been the subject of several clinical studies examining mood and psychological wellbeing.
In a randomized, double-blind, placebo-controlled study, 30 female participants consumed H. erinaceus over four weeks. Those in the active group showed significantly lower scores on the Center for Epidemiologic Studies Depression Scale (CES-D) and the Indefinite Complaints Index (ICI). Notably, subscores related to concentration, irritability, and anxiety showed trends toward improvement in the mushroom group compared to placebo.[1]
A 2025 systematic review of clinical studies on H. erinaceus supplementation found that across multiple trials, the mushroom was associated with improvements in mood, reduction in anxiety and depression symptoms, and enhanced hippocampal neurogenesis. The review noted that H. erinaceus may support elevated levels of BDNF (brain-derived neurotrophic factor) and pro-BDNF, both of which are implicated in mood regulation.[2]
The proposed mechanisms include stimulation of nerve growth factor (NGF) synthesis via hericenones derived from the fruiting body and erinacines from the mycelium, both of which may influence neuronal plasticity and resilience to psychological stressors.
Mushroom Blends and the HPA Axis: A Randomized Trial
A 2026 randomized, double-blind, placebo-controlled trial examined the effects of a medicinal mushroom blend supplement over 12 weeks in 50 participants. The intervention group showed significantly greater reductions in state anxiety as measured by both the State-Trait Anxiety Inventory (STAI-S) and the Hamilton Anxiety Scale (HAM-A) at both six and twelve weeks. Depression scores assessed via the Beck Depression Inventory (BDI) also showed significant reductions at both timepoints.
Crucially, biomarker analysis in the same study revealed significant reductions in serum cortisol and ACTH levels, alongside a decrease in CRP, a marker of systemic inflammation. These findings suggest that the observed psychological improvements may reflect measurable changes in HPA axis activity, not merely subjective perception.[3]
The study also reported improvements in fatigue measures and sleep quality, which are closely linked to stress regulation, with a trend toward increased morning melatonin levels after 12 weeks of supplementation.
Reishi and Stress-Related Fatigue
Ganoderma lucidum, commonly known as Reishi, has a long history of use in traditional East Asian medicine and is one of the most studied medicinal mushrooms. Its triterpene compounds have been examined for potential effects on the nervous system and inflammatory pathways.
A pilot clinical trial found that spore powder of G. lucidum significantly improved fatigue, quality of life, and psychological wellbeing in breast cancer patients undergoing endocrine therapy. Participants in the intervention group reported less anxiety and depression compared to controls, and the study measured significant reductions in TNF-alpha and IL-6, two cytokines that are elevated during chronic stress and are associated with depressive symptoms.[4]
While this study was conducted in a specific patient population, the inflammatory and fatigue mechanisms it addresses are relevant to general stress physiology. Research suggests that the triterpenes in Reishi may modulate inflammatory signaling pathways that overlap with stress-induced immune dysregulation.
Proposed Mechanisms in Reishi
Several pathways have been proposed to explain Reishi’s potential effects on stress-related outcomes. These include inhibition of NF-kB signaling (a key mediator of stress-induced inflammation), modulation of serotonergic and GABAergic activity in animal models, and antioxidant activity that may buffer oxidative stress associated with psychological strain. Human clinical evidence for these specific pathways remains limited, and further trials are needed.
Shared Mechanisms Across Functional Mushrooms
While each species carries distinct bioactive compounds, several mechanisms appear to be shared across functional mushrooms that may be relevant to stress and anxiety:
- Anti-inflammatory activity: Beta-glucans and polysaccharides found in Reishi, Lion’s Mane, Turkey Tail, and Cordyceps have demonstrated immunomodulatory effects that may attenuate inflammation linked to chronic stress.
- Antioxidant capacity: Oxidative stress is a well-established component of anxiety and mood disorders. Compounds in Chaga, Reishi, and Lion’s Mane have shown antioxidant activity in preclinical models.
- Neurotrophic support: Lion’s Mane is the most studied mushroom for direct neurotrophin stimulation. NGF and BDNF pathways are central to both cognitive resilience and mood regulation.
- HPA axis modulation: Some mushroom interventions have measured reductions in cortisol and ACTH, suggesting an adaptogenic effect on the stress hormone cascade.
Limitations of Current Evidence
It is important to note several limitations in the existing literature. Many studies use small sample sizes or proprietary blends that make it difficult to attribute effects to a single species. Study durations vary considerably, dosing protocols are inconsistent across trials, and most human studies focus on specific populations such as older adults or cancer patients. Extrapolating findings to general healthy adult populations should be done cautiously.
Additionally, most mechanistic data comes from in vitro or animal studies. While these provide useful hypotheses, they do not confirm clinical efficacy in humans. Functional mushrooms should not be viewed as replacements for established pharmacological or psychotherapeutic treatments for anxiety or stress disorders.

Summary
Current research suggests that certain functional mushrooms, particularly Lion’s Mane and Reishi, may support psychological wellbeing through pathways involving neurotrophic factors, HPA axis regulation, and inflammatory modulation. Randomized controlled trials have demonstrated measurable reductions in anxiety scores and stress biomarkers including cortisol and inflammatory cytokines with mushroom supplementation. However, research remains early-stage for most species, and well-powered, long-term clinical trials in healthy populations are still limited.
Individuals with anxiety or stress-related conditions should consult a qualified healthcare provider before incorporating functional mushrooms into their regimen.
